Locus Description: The androgen receptor gene is more than 90 kb long and codes for a protein that has 3 major functional domains. Migeon et al. (1981,PubMed ID:6947723) localised the androgen receptor gene between Xq13 and Xp11 by somatic cell hybridization. Wieacker et al. (1985), localized the gene between Xcen and Xq13 by RFLP. The coding sequence of the gene is divided into eight coding exons and spans a minimum of 54 kilobases. Constitutional mutations in the androgen receptor gene (AR ) impair androgen - dependent male sexual differentiation to various degrees . Somatic mutations in the AR have been found in metastatic prostate cancer. This gene contains 2 polymorphic trinucleotide repeat segments that encode polyglutamine and polyglycine tracts in the N-terminal transactivation domain of its protein. Expansion of the polyglutamine tract causes spinal bulbar muscular atrophy (Kennedy disease).
